P-50: The Effect of Sulpiride on Reproductive System in Female Rats

Background: Sulpiride is a substituted benzamide derivative and a selective dopamine D2 antagonist with antipsychotic and antidepressant activity. Sulpiride is effective in the treatment of acute and chronic schizophrenia.Sexual function is multifaceted and is affected in many ways by schizophrenia and the antipsychotic drugs used in its treatment . Sulpiride are associated with elevation of prolactin levels;hyperprolactinemia can lead to various hormonal adverse effects including sexual dysfunction,gynecomastid ,amenorrhea,and galactorrhea.. the aim of the present study was to evaluate effect of sulpiride on the ovarian functions of rat. Materials and Methods: In this study thirty adult female wister rats divided into three groups (10 per groups) .sulpiride was diluted in olive oil to provide appropriate dose for injection. Two groups of animals received 20 and 40mg/kg bw of sulpiride (ip) respectively and one group received olive oil as control group for 28 days. All rats were sacrificed by Co2 inhalation one day after last injection and the ovary was removed and fixed in10% neutural buffered formalin and processed for paraffin embedding. Five µm thick sections were stained with H&E. Light microscopical analysis was made to evaluate the structural changes induced in the ovary. Results: This study showed that sulpiride-treated females persistent diestrus resulted in unsuccessful mating and decrease pregnancy rate ,sulpiride-induced ovarian toxicity and menstrual disorders( Amenorrhea ) was seen too. Conclusion: Taking collectively, the present results highly support the idea that sulpiride has adverse effect on reproductive system of female rats.
