Abstract:

Background Egg yolk is the main cryoprotectant agent in semen freezing extenders which is used in order to protect spermatozoa against cold shock. However elimination of animal bioproducts from the cryopreservation protocol is becoming mandatory. Therefore the aim of this study is to compare a previously studied homemade soya bean lecithin based extender with a commercially available extender for ram sperm cryopreservation. MaterialsAndMethods Samples from three rams were pooled and split into two equal aliquots and diluted (1:20) with i%lecithin - 7%glycerol (L1G7) and Bioxcell®. The effects of L1G7 and Bioxcell® on sperm parameters and the in vitro fertilization ability of frozen-thawed ram spermatozoa were assessed. Results The results of this study revealed no difference between the two extenders in terms of motility viability and capacitation status. The results of in vitro fertilization in terms of rate of blastocyst formation were similar for both extenders but significantly lower than that of freshly processed ram sperm. Conclusion We conclude that both extenders are suitable for ram sperm cryopreservation.

P-127: In Vitro Assessment of Trehalose for Cryopreservation of Ram Sperm in Extender Containing Soybean Lecithin

Background: Trehalose, a non-permeable disaccharide, has a protective role related both to osmotic effect and specific interactions with membrane phospholipids, rendering hypertonic media, causing cellular osmotic dehydration before freezing, and then decreasing the amount of cell injury by ice crystallization. Within this study, an attempt was made to evaluation of two levels of 50 and 100 mM ...

Effects of Centrifugation and Different Levels of Soybean Lecithin-Based Extender on Post-Thaw Sperm Quality of Ghezel Ram Semen

The objective of this study was to specify the effects of centrifugation and different surface of soybean lecithin-based extender on post-thaw Sperm quality of Ghezel ram semen. Semen samples were gathered from 5 mature Ghezel ram two/week for 3 weeks. After initial evaluation, the approved semen samples were pooled together and split into equal parts in Falcon tubes. Three samples were combine...

Does natural honey act as an alternative to antibiotics in the semen extender for cryopreservation of crossbred ram semen?

Antibiotics are added to semen extenders to take care of heavy microbial load, however, their continuous use poses a constant threat of developing antibiotic resistance by the common microbes present in the semen. Our hypothesis was that natural honey, having antibacterial activity and rich in fructose could replace the use of antibiotics and fructose in the semen extender. Twenty-four ejaculat...

P-137: Improvement in Post-Thawed Ram Sperm Quality by Adding Cysteine in Soybean Lecithin Based Extender

Background: Ram sperm have a higher polyunsaturated fatty acids and a lower cholesterol/phospholipid ratio than other species which renders the sperm much more vulnerable to oxidative damage caused by reactive oxygen species (ROS). Oxidative stress can affects the integrity of cells at both the structural and sub structural levels. Therefore, a suitable antioxidant may prevent cryo-injuries and...
