Abstract:

Background: Sodium valproate, an anticonvulsant drug, is reported to stimulate Human Immunodeficiency Virus type 1 and Human cytomegalovirus replication. Since epileptic patients undergoing sodium valproate therapy may suffer from various virus infections, the effect of this drug on replication of viruses especially those affecting neuronal tissues such as Herpes simplex virus type 1 is worthy of investigation. Methods: Viral replication was studied by quantal response method. Electron microscopy was also performed on cell monolayers treated with the drug and infected with the virus. Results: Significant reduction in viral infectivity was observed in cell cultures exposed to 0.5-2 mM of sodium valproate either one hr before or after infection. The corresponding electron microscopic examination revealed a very few intracytoplasmic enveloped virions and a marked reduction in the number of intracytoplasmic nucleocapsids in drug-treated virus infected cells compared with those of infected control. However, significant stimulation of virus replication was found upon treatment of cells with 1 mM (p<0.01) and 2 mM (p<0.001) of the drug 24 hours prior to infection. No remarkable change was seen in corresponding electron micrographs. Conclusion: Although the ultimate outcome of this study awaits in vivo assessments, possible stimulation of Herpes simplex virus type 1 replication by sodium valproate should be considered by clinicians prescribing this drug.Iran J Med Sci 2006; 31(1): 28-32. Keywords ● HSV-1 ● Sodium valproate ● inhibition ● electron microscopy

Effects of Morphine on Replication of Herpes Simplex Virus Type 1&2

Background and Aims: Several drugs are being used in treatment of HSV infection in human but still introducing an effective safe drug is desirable. Methods: We investigated the inhibitory effect of morphine on replication of HSV in vitro. Results: The results indicated that a concentration of up to 200 ug/ml morphine had a limited effect on Vero cell viability. At this concentration the growt...

A Preliminary Study on the Molecular Identification of Herpes Simplex Virus Type 1 in Iranian Population

Herpes Simplex Virus Type 1 (HSV-1) is a member of the Herpesviridae family that causes herpetic disease in human. During the last 25-30 years, many investigations have been conducted on pathogenesis and molecular biology of this virus in several countries. In Iran HSV-1 has been isolated from patients and detected using immunological techniques. In this study, we investigated the molecular asp...

A review study on the effect of Iranian herbal medicines against in vitro replication of herpes simplex virus

Objective: There are a number of published data indicating in vitro anti-HSV activity of some of Iranian herbal extracts with no systematic review to discuss these results. Therefore, this article was aimed to review and discuss the methods carried out and the phytochemistry and bioactivity of the extracts used and also conclusions provided in these publications. Materials and Methods: Publishe...

The Changing Epidemiology of Herpes Simplex Virus Type 1 Infection: The Associated Effects on the Incidence of Ocular Herpes

Herpes simplex virus type 1 (HSV-1) with a worldwide distribution has been reported in all human populations, resulting in a clinical spectrum of infections. Although HSV type 2 (HSV-2) is known as the most common cause of genital herpes, an increasing number of cases with genital herpes are caused by HSV-1. The present study aimed to discuss the changes in the epidemiology of HSV-1 infection i...

Replication Characteristics of Herpes Simplex Virus Type-1 (HSV-1) Recombinants in 3 Types of Tissue Cultures

A complication in the analysis of the role of ICP34.5 gene in the herpes simplex virus type-1 (HSV-1) lifecycle is the presence of overlapping antisense gene, open reading frame P (ORF P), which is also deleted in HSV-1 ICP34.5 negative mutants. A HSV-1 wild type strain (17+) ICP34.5/ORF P deletion mutant (1716) is totally avirulent in animal models and impaired in a number of in vitro function...
