CD44 expression changes and increased apoptosis in MCF-7 cell line of breast cancer in simulated microgravity condition

Authors

Abstract:

Introduction: Studies have shown that simulated microgravity (SMG) affects tumor cell proliferation and metastasis. However, the underlying mechanism and its molecular basis are still not well known. In recent years, due to the role of CD44 in breast cancer and its high expression in invasive basal tumors, it has been the subject of extensive research. There is a conflicting data on the role of the CD44 in both tumor suppression and promotion. The aim of present study was to investigate the gene expression of CD44 and apoptosis in MCF-7 cell line of breast cancer in simulated microgravity condition. Materials and Methods: MCF-7 was proliferated under normal gravity and microgravity (1 and 3 days) using clinostat. Gene expression and apoptosis was measured using real-time PCR technique and Annexin V staining kit. Results: The results indicated that SMG reduced gene expression (50%) after 1 day and increased it (four times) during 3 days. Also, apoptosis was increased 40% after 1 day. Conclusion: It seems the response of cancer cells to microgravity is time dependent and microgravity treatment for 1 day may have a positive effect on cancer characteristics for achievement to effective cancer therapy.

Induction of Apoptosis and Non-Apoptosis in Human Breast Cancer Cell Line (MCF-7) by Cisplatin and Caffeine

Background: Molecular targeted therapy by different cell death inducers are recently considered in cancer therapy. The aim of this study was to compare the effect of cisplatin and inositol trisphosphate kinase inhibitor (caffeine) on human breast cancer cell line (MCF-7). The pattern of cell death in MCF-7 cells following the exposure to cisplatin and caffeine in individual and combination form...

Growth Inhibition and Apoptosis Induction of Salvia chloroleuca on MCF-7 Breast Cancer Cell Line

Fragrant species of the genus Salvia have been attributed many medicinal properties, which include anticancer activity. In the present study, cytotoxic properties of total methanol extract of Salvia chloroleuca Rech. f. & Aellen and its fractions were investigated on MCF-7, a breast carcinoma cell line. Malignant and non-malignant cells were cultured in RPMI medium and incubated with different ...

Effect of Silibinin on Maspin and ERα Gene Expression in MCF-7 Human Breast Cancer Cell Line

Background and objective: According to reports, a serine protease inhibitor (Maspin) suppresses metastasis, invasion and angiogenesis in breast and prostate cancers. Silibinin is a natural polyphenolic flavonoid with anti-cancer activity. We assessed the effects of silibinin on cell viability, maspin and ERα gene expression in MCF-7 cell line. <s...

The Activity of Sirtuin 1 in MCF-7 Breast Cancer Cell Line: The Effects of Visfatin

Background & Objectives: Breast cancer is the most common cancer and the second leading cause of cancer deaths among women. Obesity, hormones, and growth factors are the risk factors for this kind of cancer. One of the changes observed in patients suffering from breast cancer is the elevated Visfatin or nicotinamide phosphoribosyl transferase (NAMPT) in their tumor tissues and blood. The increa...
