چکیده
Background: Olfactomedin 1 (Olfm1) is a highly conserved secreted glycoprotein with the unknown mechanism of action. Results: Olfm1 binds to NgR1 and its binding reduced binding of NgR1 co-receptors, p75NTR and LINGO-1. Conclusion: Olfm1 is a novel NgR1 ligand that modulates the functions of the NgR1 complex in axonal growth. Significance: Olfm1 may be used to facilitate neuronal growth after axonal damage.

Olfactomedin 1 interacts with the Nogo A receptor complex to regulate axon growth.
Olfm1, a secreted highly conserved glycoprotein, is detected in peripheral and central nervous tissues and participates in neural progenitor maintenance, cell death in brain, and optic nerve arborization. In this study, we identified Olfm1 as a molecule promoting axon growth through interaction with the Nogo A receptor (NgR1) complex. Olfm1 is coexpressed with NgR1 in dorsal root ganglia and re...
متن کاملA multi-domain fragment of Nogo-A protein is a potent inhibitor of cortical axon regeneration via Nogo receptor 1.
Nogo-A limits axon regeneration and functional recovery after central nervous system injury in adult mammals. Three regions of Nogo-A (Nogo-A-24, Nogo-66, and Nogo-C39) interact with the neuronal Nogo-66 receptor 1 (NgR1). Nogo-66 also interacts with a structurally unrelated cell surface receptor, paired immunoglobulin-like receptor (PirB). We show here that the other two NgR1-interacting domai...

Neurons in the peripheral nervous system (PNS) have the capability to regenerate after injury or disease while central nervous system (CNS) neurons do not. Elucidation of the reasons for this difference in regenerative ability is crucial to developing treatments for sufferers of CNS disorders, injury, and stroke. Therefore, my lab investigates molecular mechanisms underlying neuronal repair, ax...
متن کاملNogo-A interacts with the Nogo-66 receptor through multiple sites to create an isoform-selective subnanomolar agonist.
Nogo is a myelin-derived protein that limits axonal regeneration after CNS injury. A short hydrophilic Nogo-66 loop between two hydrophobic domains of Nogo binds to a Nogo-66 receptor (NgR) to inhibit axonal outgrowth. Inhibition of axon outgrowth and cell spreading by a second Nogo domain, termed Amino-Nogo-A, is thought to be mediated by a distinct receptor complex. Here, we define a novel No...

Retinoic acid keeps Lingo-1 quiet R etinoic acid (RA) promotes axon regeneration by silencing a key inhibitory receptor, Puttagunta et al. report. Neuronal regrowth after injury is limited, in part, by inhibitory proteins released upon disruption of the surrounding myelin sheath. These proteins bind to the Nogo receptor complex on the neuronal plasma membrane, thereby activating the RhoA GTPase...
