چکیده

The use as an embedding resin for ultrathin sectioning of a cross-linked triple copolymer of N-vinylpyrrolidone, acrylonitrile, and ethylene glycol dimethacrylate is described. The first of these components is miscible with water, in all proportions, and can be used as a dehydrating agent, or, alternatively, ethanol may be used in the standard way. Polymerization is carried out at 37 degrees C or even lower temperatures. This resin is unsuitable for use after osmium fixatives, but after permanganates it gives results similar to epoxy. Photographs of rye root-tip cells fixed in permanganate and sectioned in this resin are presented. Because of the water-permeable nature of the product and low polymerization temperature, this resin appears to have possibilities for histochemistry.

Araldite as an Embedding Medium for Electron Microscopy

Epoxy resins are suitable media for embedding for electron microscopy, as they set uniformly with virtually no shrinkage. A mixture of araldite epoxy resins has been developed which is soluble in ethanol, and which yields a block of the required hardness for thin sectioning. The critical modifications to the conventional mixtures are the choice of a plasticized resin in conjunction with an alip...

A New Maraglas, D.e.r.® 732, Embedment for Electron Microscopy

A new epoxy embedding mixture for biological material recently has been introduced by Freeman and Spurlock (1). This mixture consists of Mara-glas 655, Cardolite NC-513, dibutyl phthalate, and the curing agent benzyldimethylamine (BDMA). Maraglas has certain advantages over other resins and epoxies, notably, wide range of miscibility, low viscosity, ease of sectioning, good staining qualities, ...

A new and simple cryosectioning protocol for pollen analysis under light microscopy

Conventional plant microtechniques typically involve fixation, dehydration, and embedding in acrylic or epoxy resins followed by room temperature microtomy. Some cryomethods have been used for pollen grain analysis under light microscopy. However, these protocols do not allow for obtaining sections with similar or better quality compared to resin embedding methods. In this paper, a simple cryop...

A New Epoxy Embedment for Electron Microscopy

A new epoxy embedding mixture has been developed utilizing Maraglas 655 and Cardolite NC-513 with benzyldimethylamine (BDMA) as a curing agent. This epoxy mixture permits cellular preservation comparable to that obtained with Epon 812, ease of preparation of tissues, a wide range of miscibility, low viscosity, and, most important, ease of sectioning on a Porter-Blum microtome. In contrast to Ep...

Microinjection of Xenopus Laevis Oocytes

Microinjection of Xenopus laevis oocytes followed by thin-sectioning electron microscopy (EM) is an excellent system for studying nucleocytoplasmic transport. Because of its large nucleus and high density of nuclear pore complexes (NPCs), nuclear transport can be easily visualized in the Xenopus oocyte. Much insight into the mechanisms of nuclear import and export has been gained through use of...
