A Reproducible Standard Substrate in Starch-amylase Viscosimetry
نویسندگان
چکیده
A viscosimetric method of estimating enzyme concentration with special reference to amylase has been discussed in another communication 1 from this laboratory; and, in a later report, * the effects of variation in the concentration of starch from the standard lot and variation in lot of prepared soluble starch have been contrasted, indicating that only within a small range of variation of p (defined there as the relative viscosity of the blank with respect to water) may comparability of digestion rates be expected. These experiments were completed in June, 1930, but were not published at that time. The object was to demonstrate some of the difficulties (a subject treated also by Chesley) 3 which may be encountered in attempts to compare results with different lots of soluble starch in the hope that some method of obtaining reproducible standard substrate solutions might be suggested. In the present communication we propose to outline a method of approximating the 3 per cent starch solution previously suggested as a standard 2 by use of a composite of other lots of soluble starch. The experimental procedure involved simply the mixing of soluble starch of two original lots (one yielding a more viscous and the other a less viscous solution than the standard, if preparation was made in standard manner I, ~) in such proportion as to give a composite approximating the standard with respect to the value of p obtained from the conventional blank, 1 part of 0.85 per cent NaC1 added to 5 parts by volume of a 3 per cent starch solution prepared in the standard manner, at 37.50 ±0.03°C. t Thompson, W. A Study of the Effects of Certain Variations in Preparation of a Starch Substrate in Amylase Viscosimetry
The results of employing different concentrations of starch and starch from different lots as substrates in the viscosimetric estimation of amylase concentration have been compared with the object of ultimate attainment of reproducible standards, independent of successive comparison.
متن کاملStudies in Starch Amylase Viscosimetry
The viscosimetric method of estimating enzyme concentration was first developed by Northrop and Hussey (1). In studies of the effect of irradiation of trypsin and pepsin solutions by Hussey and Thompson (2) certain modifications of the original methods were employed in order to decrease the opportunity for personal influence in curve plotting, as in extrapolation for the initial viscosity of th...
متن کاملKinetic nephelometric procedure for measurement of amylase activity in serum.
We developed a kinetic procedure for determination of amylase activity in serum by use of nephelometric measurements. Light scattering from the substrate, a stable suspension of starch, is decreased as amylase hydrolyzes the starch to soluble fragments. Values obtained for serum amylase correlate closely with those determined by a method in which a starch-dye complex is used. Precision of the n...
متن کاملThe Role of Highly Conserved Tryptophan in the Sixth Conserved Region at Substrate Specificity of α- amylase
Early in this study, an α-Amylase from Bacillus megaterium WHO (BMW) was isolated from hot springs of Ramsar (North of Iran), and its gene was cloned in E.coli. Based on its conserved sequence regions and substrate specificity, it was classified as intermediary group enzymes with the specificity of oligo-1,6-glucosidase and neopullulanase subfamilies. In the sixth conserved re...
متن کاملEffect of temperature , pH , substrate ( Starch ) and glucose on stability of α - amylase from Bacillus licheniformis ATCC 6346
Thermostable α-amylases (1,4-α-D-glucan glucanohydrolase; E.C.3.2.1.1) from Bacillus species are of great industrial importance in the production of corn syrup or dextrose. The present study was aimed to determine the effect of temperature, pH, substrate and glucose on stability of α-amylase from Bacillus licheniformis ATCC 6346. To determine the effect of temperature and pH on the stability of...
