چکیده

A 426 bp cDNA encoding a predicted small heat shock protein (smHSP) was isolated from Wuchereria bancrofti lambda zap L3 cDNA expression library by immunoscreening with microfilaremic sera. The open reading frame of the cDNA clone encodes a predicted protein of 142 amino acids (aa), which had high sequence identity with other nematode smHSPs. The homologous regions conserved in several different nematode species reflect its importance in parasites that require mammalian host as a part of their development. SmHSPs and alpha-crystallin constitute a family of related molecular chaperons that exhibit striking variability in size ranging from 10 to 43 kDa. Here we describe the cloning and characterization of this cDNA clone encoding predicted 15.5 kDa smHSP of W. bancrofti.

Cloning and Expression of Heat Shock Protein 60kDa Gene from Brucella melitensis as Subunit Vaccine

Brucellosis is caused by the bacterium Brucella and affects various domestic and wild species. GroEL (Heat Shock Protein 60kDa) as one of the major antigens that stimulate the immune system, increases Brucella survival. The aim of the current study was to clone and express GroEL in Escherichia coli in order to design subunit vaccine. Amplifying was performed using specific primers. The full-len...

Cloning and characterization of a potentially protective chitinase-like recombinant antigen from Wuchereria bancrofti.

While there is no direct evidence demonstrating the existence of protective immunity to Wuchereria bancrofti infection in humans, the presence of individuals, in populations in areas where infection is endemic, with no clinical evidence of past or current infection despite appreciable exposure to the infective larvae, suggests that protective immunity to filarial parasites may occur naturally. ...

Expression of Recombinant Heat-Shock Protein 70 of MCAN/IR/96/LON-49, a Tool for Diagnosis and Future Vaccine Research

Background: Heat shock protein 70 (HSP70) is present in all organisms studied so far, and is a major immunogen in infections caused by pathogens including Leishmania spp. Objective: The aim of this study was to clone and express HSP70 from L. infantum strain MCAN/IR/96/LON-49 and evaluate antibody response against HSP70 in visceral leishmaniasis (VL). Methods: The L. infantum HSP70 gene segment...

Onchocerca volvulus heat shock protein 70 is a major immunogen in amicrofilaremic individuals from a filariasis-endemic area.

Infestation with organisms causing lymphatic filariasis (i.e. Wuchereria bancrofti and Brugia malayi) results in a variety of clinical presentations. It is possible that some of the variation is due to differences in host response to parasite. To determine whether individuals who live in an endemic area but differ in their clinical manifestations respond to different filarial antigens, we scree...

Cloning and Characterization of cbhII Gene fromTrichoderma parceramosum and Its Expressionin Pichia pastoris

The genomic and cDNA clones encoding cellobiohydrolase II (CBHII) have been isolated and sequenced from a native Iranian isolate of Trichoderma parceramosum, a high cellulolytic enzymes producer isolate. This represents the first report of cbhII gene from this organism. Comparison of genomic and cDNA sequences indicates this gene contains three short introns and also an open reading frame codin...
