چکیده
We evaluated the effectiveness of serological and parasitological methods for cutaneous leishmaniasis (CL) diagnosis in patients from the central region of Paraná state, southern Brazil. Five groups were compared: clinical diagnosis, parasitological diagnosis, communicants, inhabitants of a non-endemic area and carriers of other etiologies. Two antigens were prepared from promastigotes of Leishmania (Viannia) braziliensis and Leishmania (Leishmania) amazonensis for indirect immunofluorescence assay, ELISA and immunoblotting. The parasitological approaches detected 79.3% of the patients with a clinical diagnosis; the parasites were identified by PCR as L. (V.) braziliensis. Serological methods showed 95% sensitivity for homologous antigens. Immunoblotting revealed specific proteins for diagnosis of CL and detected 96.6% of the patients when L. (V.) braziliensis was used as an antigen, and 83.3% with L. (L.) amazonensis. This study demonstrated the importance of differential diagnosis for leishmaniasis; the association of two or more indirect methods increased diagnosis sensitivity.

American cutaneous leishmaniasis (ACL) is an endemic disease in northern Paraná State, where it affects humans and dogs. This study aimed to verify the occurrence of the canine disease in ACL endemic areas. From September 1999 to July 2002, dogs were investigated in 7 rural areas of 5 municipalities where autochthonous human cases of ACL had been reported. Parasitological and serological method...
متن کاملDiagnosis of American cutaneous leishmaniasis by enzyme immunoassay in patients from Northern Paraná State, Brazil.
American cutaneous leishmaniasis (ACL) is an endemic disease caused by Leishmania parasites. The ACL diagnosis is commonly accomplished by parasitological and immunological methods. The objective of this work was to evaluate the enzyme immunoassay (EIA) for ACL laboratory diagnosis. IgG antibodies against Leishmania (Viannia) braziliensis promastigotes were researched. For the method standardiz...
متن کاملA Comparison Study between Parasitological and Molecular Methods for Diagnosis of Acute and Chronic Anthroponotic Cutaneous Leishmaniasis
Introduction: Anthroponotic cutaneous leishmaniasis (ACL) is still a major public health problem in the northeast and central parts of Iran. This study was designed to compare microscopy and cultivation methods with PCR amplification of kinetoplast DNA and ITS1 followed by RFLP analysis for diagnosis of acute and chronic ACL. Methods: In this study, 66 patients with ACL including 24 acute and 4...
متن کاملThe Comparison of Direct Microscopic Examination and Culture Results in Diagnosis of Cutaneous Leishmaniasis
Abstract Background and Objectives: one of the endemic foci for Cutaneous Leishmaniasis in Iran is Mashhad in which limited outbreak have recently been reported. The commonly used method for diagnosis is the clinical features confirmed by direct microspic examination and culture or biopsy. We compared these two tests to determine the level of their sensitivity, specificity and positive predicti...
متن کاملComparison of fine needle aspiration and direct skin smear in the diagnosis of acute cutaneous leishmaniasis: polymerase chain reaction as a reference method
Background: Accurate diagnosis of cutaneous leishmaniasis wouldavoid unnecessary treatment and scar formation. Direct smear isthe most common method for the diagnosis of this disease but itsnegative result could not rule out the infection; so, the need formore sensitive methods is obvious. We conducted this study tocompare the efficiency of direct skin smear with smears preparedby fine needle a...
