The effect of viomycin on elongation steps in cell-free polypeptide synthesis.

چکیده
that VMinhibits protein synthesis was first shown in intact Mycobacterium aviurrP and it is a ribosomal inhibitor since VM resistance of certain mutants of Mycobacterium smegmatis and Mycobacterium tuberculosis can be attributed to an alteration of ribosomes4~7). The characteristics of VMbinding to ribosomes have been studied. The drug reduced the amounts of [3H]dihydrostreptomycin bound to ribosomes of M. smegmatis and Escherichia coli suggesting an interaction between the binding sites of these antibiotics8). Kinetics of initiating polypeptide elongation in an IRES-dependent system
The intergenic IRES of Cricket Paralysis Virus (CrPV-IRES) forms a tight complex with 80S ribosomes capable of initiating the cell-free synthesis of complete proteins in the absence of initiation factors. Such synthesis raises the question of what effect the necessary IRES dissociation from the tRNA binding sites, and ultimately from all of the ribosome, has on the rates of initial peptide elon...
متن کاملMolecular mechanism of viomycin inhibition of peptide elongation in bacteria.
Viomycin is a tuberactinomycin antibiotic essential for treating multidrug-resistant tuberculosis. It inhibits bacterial protein synthesis by blocking elongation factor G (EF-G) catalyzed translocation of messenger RNA on the ribosome. Here we have clarified the molecular aspects of viomycin inhibition of the elongating ribosome using pre-steady-state kinetics. We found that the probability of ...
متن کاملColicin E3 cleavage of 16S rRNA impairs decoding and accelerates tRNA translocation on Escherichia coli ribosomes
The cytotoxin colicin E3 targets the 30S subunit of bacterial ribosomes and specifically cleaves 16S rRNA at the decoding centre, thereby inhibiting translation. Although the cleavage site is well known, it is not clear which step of translation is inhibited. We studied the effects of colicin E3 cleavage on ribosome functions by analysing individual steps of protein synthesis. We find that the ...
متن کاملAction of streptothricin F on ribosomal functions.
The effect of streptothricin F on elongation factor-dependent and on elongation factor-free translation systems was studied. Streptothricin F inhibits factor-dependent as well as factor-free polypeptide synthesis. The results suggest that streptothricin F inhibits polypeptide synthesis via interaction with the ribosome. In partial reactions streptothricin F impairs EF-G-dependent translocation ...
متن کاملSnapShot: Antibiotic Inhibition of Protein Synthesis I
Elongation (translocation) Most aminoglycoside antibiotics induce translational misreading by promoting binding of near-cognate tRNAs, but the biological effect is probably due to inhibition of the translocation reaction and promotion of back-translocation. Edeine Edeine A Initiation (fMet-tRNA binding) Edeine prevents binding of the initiator tRNA to the 30S subunit in an mRNA-dependent manner...
