Monoclonal anti-mouse laminin antibodies: AL-1 reacts with laminin alpha1 chain, AL-2 with laminin beta1 chain, and AL-4 with the coiled-coil domain of laminin beta1 chain. Access to the published version may require journal subscription. Abstract We analyzed the reactivity of three different commercially available rat monoclonal antibodies raised against mouse laminin-α1β1γ1 (laminin-111), AL-1, AL-2, and AL-4. Using ELISA assays, Western blot analysis and immunostainings we present refined epitope maps for these three laminin monoclonals. AL-1 reacted, as predicted with laminin α1 chain. AL-4 has also been marketed as an α1 chain specific probe, but we show here that AL-4 detects mouse laminin β1 chain, in the distal part of the coiled-coil region. AL-2 was predicted to react with all three chains near the cross region, but seems to primarily react with laminin β1 chain.

Crystal Structure of the Heterotrimeric Integrin-Binding Region of Laminin-111
Laminins are cell-adhesive glycoproteins that are essential for basement membrane assembly and function. Integrins are important laminin receptors, but their binding site on the heterotrimeric laminins is poorly defined structurally. We report the crystal structure at 2.13 Å resolution of a minimal integrin-binding fragment of mouse laminin-111, consisting of ∼50 residues of α1β1γ1 coiled coil ...

Interaction of epithelial and endothelial cells with basement membranes plays a critical role in induction and maintenance of their differentiation phenotypes. The biological activities of basement membranes can in large part be attributed to laminins, the major glycoproteins of basement membranes throughout the vertebrate body. Laminins are disulfide-linked heterotrimers of three distinct but ...
متن کاملLaminin-121--recombinant expression and interactions with integrins.
Laminin-121, previously referred as to laminin-3, was expressed recombinantly in human embryonic kidney (HEK) 293 cells by triple transfection of full-length cDNAs encoding mouse laminin α1, β2 and γ1 chains. The recombinant laminin-121 was purified using Heparin-Sepharose followed by molecular sieve chromatography and shown to be correctly folded by electron microscopy and circular dichroism (...
متن کاملDrosophila laminin A chain sequence, interspecies comparison, and domain structure of a major carboxyl portion.
Recent studies ascribed some biological actions of cell adhesion and cell outgrowth to the carboxyl-most 1200 amino acids of vertebrate laminin A chains. Here we report a 6.1-kilobase pair nucleotide cDNA sequence encoding 1951 amino acids and the carboxyl end of a Drosophila laminin A chain. It corresponds to the mouse laminin A domains G, I, II, and III, but may represent a different type of ...
متن کاملNative Chick Laminin-4 Containing the 132 Chain (s-Laminin) Promotes Motor Axon Growth
After denervation of muscle, motor axons reinnervate original synaptic sites. A recombinant fragment of the synapse specific laminin 132 chain (s-laminin) was reported to inhibit motor axon growth. Consequently, a specific sequence (leucine-arginineglutamate, LR E) of the laminin 132 chain was proposed to act as a stop signal and to mediate specific reinnervation at the neuromuscular junction (...
