چکیده

Background The current tools available for diagnosing food allergy include the clinical history, trial elimination diets, diet diaries, skin prick testing (SPT) and allergen-specific serum IgE testing. Since 15 years, development of allergen-based microarrays using recombinant/natural proteins is increasing, resulting in commercially available microarrays to detect specific IgE against more than 100 allergenic compounds. For the moment, the number of available recombinant allergens and the cost of their development limit the enhancement of allergen microarray technology. The goal of our study is to explore and optimize allergen-microarray from food allergen extracts and trying to create a low-cost IgE detecting platform.

Evidence for higher sensitivity of recombinant Tri a 36 compared to omega-5-gliadin for diagnosis of wheat food allergy

Background Wheat is one of the most important food allergen sources. Using natural wheat allergen extracts for serological diagnosis of wheat-induced food allergy false positive test results are frequently obtained, in particular in grass pollen allergic patients. Therefore, Tri a 19, an omega-5-gliadin, which is known as a major allergen in wheat dependent exercise induced anaphylaxis (WDEIA) ...

Diagnosis of food allergies: the impact of oral food challenge testing

A diagnosis of food allergies should be made based on the observation of allergic symptoms following the intake of suspected foods and the presence of allergen-specific IgE antibodies. The oral food challenge (OFC) test is the most reliable clinical procedure for diagnosing food allergies. Specific IgE testing of allergen components as well as classical crude allergen extracts helps to make a m...

Simultaneous quantification of multiple specific food allergen proteins indicates varied allergen content in diagnostic and therapeutic preparations

In order to help allergic patients manage often severe symptoms, food manufacturers are required to list allergens on labels whereas researchers are working to improve diagnostics and develop effective immunotherapies. Existing tools using generic immunoassays do not provide precise identification or quantification of specific food allergens. Our goal was to develop an accurate, sensitive and h...

A Novel Multipeptide Microarray for the Specific and Sensitive Mapping of Linear IgE-Binding Epitopes of Food Allergens.

BACKGROUND The identification of B-cell epitopes of food allergens can possibly lead to novel diagnostic tools and therapeutic reagents for food allergy. We sought to develop a flexible, low-tech, cost-effective and reproducible multipeptide microarray for the research environment to enable large-scale screening of IgE epitopes of food allergens. METHODS Overlapping peptides (15-mer, 4 amino ...

Recombinant Allergens in Structural Biology, Diagnosis, and Immunotherapy.

The years 1988-1995 witnessed the beginning of allergen cloning and the generation of recombinant allergens, which opened up new avenues for the diagnosis and research of human allergic diseases. Most crystal and solution structures of allergens have been obtained using recombinant allergens. Structural information on allergens allows insights into their evolutionary biology, illustrates clinic...
