Serotonin, cytokines, p11, and depression.

چکیده
F or at least four decades, the field of psychoimmunology has attracted numerous investigators based on diverse hints linking the immune system to emotional behavior. The administration of cytokines for the treatment of various diseases is often associated with depression, which frequently accompanies autoimmune disease. Now, in PNAS, Greengard and colleagues (1) report a specific molecular pathway linking cytokines and the actions of antidepressant drugs. Their findings confound established wisdom, as they imply that brain cytokines exert antidepressant actions and mediate the influences of the principal serotonin-specific reuptake inhibitor (SSRI) antidepressant drugs. How the present discoveries fit into the context of our understanding of antidepressant actions can be summarized as follows.

Alterations in 5-HT1B receptor function by p11 in depression-like states.
The pathophysiology of depression remains enigmatic, although abnormalities in serotonin signaling have been implicated. We have found that the serotonin 1B receptor [5-hydroxytryptamine (5-HT1B) receptor] interacts with p11. p11 increases localization of 5-HT1B receptors at the cell surface. p11 is increased in rodent brains by antidepressants or electroconvulsive therapy, but decreased in an ...

p11 (S100A10), a member of a large family of S100 proteins, interacts with serotonin receptor 1B (5-HTR1B), modulates 5-HT1B receptor signal transduction, and is required for antidepressant responses to activation of this receptor. In the current study, we investigated the specificity of the interaction between 5-HTR1B and p11 by screening brain-expressed S100 proteins against serotonin and nor...

Depression is a mood continuum, ranging from normal but temporary "bad days" to a completely disabling clinical condition characterized by overwhelming despair that lasts for weeks or months. A popular theory is that a breakdown in signaling by the brain neurotransmitter serotonin (5-hydroxytryptamine; 5-HT) is critically involved in the symptoms of clinical depression, but the nature of this d...
متن کاملProbable involvement of p11 with interferon alpha induced depression.
Depression is one of the major side effects of interferon alpha (IFN-α) treatment, but the molecular mechanism underlying IFN-α-induced depression remains unclear. Several studies have shown that the serotonin receptors 5-HTR1b and 5-HTR4 play key roles in the anti-depression effects associated with p11 (S100A10). We investigated the effects of IFN-α on the regulation of p11, 5-HTR1b and 5-HTR4...
متن کاملAntidepressant treatment is associated with epigenetic alterations in the promoter of P11 in a genetic model of depression.
P11 (S100A10) has been associated with the pathophysiology of depression both in human and rodent models. Different types of antidepressants have been shown to increase P11 levels in distinct brain regions and P11 gene therapy was recently proven effective in reversing depressive-like behaviours in mice. However, the molecular mechanisms that govern P11 gene expression in response to antidepres...
