چکیده

OBJECTIVE To investigate the effect of troglitazone, a proliferator-activated receptor-γ (PPARγ) agonist, on the expressions of intercellular adhesion molecule-1 (ICAM-1) and matrix metalloproteainse-9 (MMP-9) and cell proliferation in HeLa cells. METHODS MTT assay was used to measure the cell viability at different time points (0, 24, 48 and 72 h) after exposure to troglitazone. RT-PCR and Western blotting were employed to detect the mRNA and protein expressions of ICAM-1, MMP-9 and PPARγ in the cells. Electrophoretic mobility shift assay (EMSA) was performed to assess the changes in DNA binding activity of PPARγ. RESULTS The viability of HeLa cells were time-dependently inhibited by troglitazone, which also significantly reduced the mRNA and protein expressions of ICAM-1 and MMP-9 and increased PPARγ expression. The effects of troglitazone in inhibiting the cell viability and reducing ICAM-1 and MMP-9 expressions were antagonized by the application of the PPARγ antagonist GW9662. The result of EMSA also showed significantly increased DNA binding activity of PPARγ in the cells exposed to troglitazone. CONCLUSION The PPARγ agonist troglitazone can inhibit the expression of ICAM-1 and MMP-9 in HeLa cells by up-regulating PPARγ.

Telmisartan exerts anti-tumor effects by activating peroxisome proliferator-activated receptor-γ in human lung adenocarcinoma A549 cells.

Telmisartan, a member of the angiotensin II type 1 receptor blockers, is usually used for cardiovascular diseases. Recent studies have showed that telmisartan has the property of PPARγ activation. Meanwhile, PPARγ is essential for tumor proliferation, invasion and metastasis. In this work we explore whether telmisartan could exert anti-tumor effects through PPARγ activation in A549 cells. MTT a...

Troglitazone Inhibits Matrix Metalloproteinase-9 Expression and Invasion of Breast Cancer Cell through a Peroxisome Proliferator-Activated Receptor γ-Dependent Mechanism

Purpose Peroxisome proliferator-activated receptor γ (PPARγ) is involved in the pathology of numerous diseases including atherosclerosis, diabetes, obesity, and cancer. Matrix metalloproteinases (MMPs) play a significant role in tissue remodeling related to various processes such as morphogenesis, angiogenesis, tissue repair, invasion, and metastasis. We investigated the effects of PPARγ on MMP...

Peroxisome-proliferator-activated receptor γ suppresses Wnt/β-catenin signalling during adipogenesis

The Wnt/β-catenin signalling pathway appears to operate to maintain the undifferentiated state of preadipocytes by inhibiting adipogenic gene expression. To define the mechanisms regulating suppression of Wnt/β-catenin signalling, we analysed the βcatenin expression in response to activation of transcription factors that regulate adipogenesis. The results show an extensive down-regulation of nu...

Inhibition of janus kinase 2 by compound AG490 suppresses the proliferation of MDA-MB-231 cells via up-regulating SARI (suppressor of AP-1, regulated by IFN)

Objective(s): The Janus kinase-signal transducers and activators of transcription signaling pathway (JAK/STAT pathway) play an important role in proliferation of breast cancer cells. Previous data showed that inhibition of STAT3 suppresses the growth of breast cancer cells, but the associated mechanisms are not well understood. This study aims to investigate the effect and associated mechanisms...

Standardized Punica Granatum Pericarp Extract, Suppresses Tumor Proliferation and Angiogenesis in a Mouse Model of Melanoma: Possible Involvement of PPARα and PPARγ Pathways

Melanoma is a challenging disease to treat. Punica granatum L. has a potential anticancer effect. This study determined the antiproliferative and antiangiogenic potential of the extract from pomegranate peel (PPE) in melanoma. Melanoma cells (1 × 106) were injected to C57BL6 mice subcutaneously. On 8th day, mice were randomly divided into 9 groups. Group 1 was considered as control and received...
