Reinitiation of DNA synthesis in senescent human fibroblasts upon fusion with cells of unlimited growth potential
نویسندگان
چکیده
Postreplicative, "senescent" human fibroblasts were fused to HeLa or to SV-40 transformed human fibroblasts with Sendai virus. DNA synthesis was reinitiated in senescent nuclei in a high proportion of the heterodikaryons. The [3H]thymidine labeling index of senescent fibroblast nuclei in heteropolykaryons was a function of the ratio of HeLa to senescent nuclei.

Evidence for endogenous polypeptide-mediated inhibition of cell-cycle transit in human diploid cells
Previous studies have shown that the senescent phenotype is dominant with respect to DNA synthesis in fusions between late passage and actively replicating human diploid fibroblasts. Brief postfusion treatments with the protein synthesis inhibitor cycloheximide (CHX) or puromycin have been found to significantly delay (by 24-48 h) the inhibition of entry into DNA synthesis of young nuclei in he...
متن کاملSupportive Effects of Human Embryonic Fibroblast Cell Lines on Growth and Proliferation of EBV-Transformed Lymphoblastoid Cells
Human diploid fibroblast cells produce a spectrum of necessary growth factors and extracellular matrix (ECM) components essential for growth and proliferation of a variety of other cell types. In this study, the effect of five human embryonic fibroblast cell lines, isolated from liver, lung, skin and foreskin tissues, was investigated. A coculture system analyse was employed to cloning efficien...
متن کاملSenescent phenotype can be reversed by reduction of caveolin status.
Hyporesponsiveness to growth factors is one of the fundamental characteristics of senescent cells. We previously reported that the up-regulation of caveolin attenuates the growth factor response and the subsequent downstream signal cascades in senescent human diploid fibroblasts. Therefore, in the present experiment, we investigated the modulation of caveolin status in senescent cells to determ...
متن کاملSenescent human fibroblasts increase the early growth of xenograft tumors via matrix metalloproteinase secretion.
Although cellular senescence is believed to have a tumor suppressor function, senescent cells have been shown to increase the potential for growth of adjacent cancer cells in animal models. Replicatively senescent human fibroblasts increase the growth of cotransplanted cancer cells in vivo, but the role of cells that have undergone damage-mediated stress-induced premature senescence (SIPS) has ...
