چکیده

The type-III interferon (IFN) family is composed of 3 molecules in humans: IFN1 (interleukin-29 [IL-29]), IFN2 (IL-28A), and IFN3 (IL-28B), each of which signals through the same receptor complex. Plasmacytoid dendritic cells (pDCs) are major IFNproducers among peripheral lymphocytes. Recently, it has been shown that IFN1 exerts a powerful inhibitory effect over the T-helper 2 (Th2) response by antagonizing the effect of IL-4 on CD4 T cells and inhibiting the production of Th2-associated cytokines. Here, we asked whether Th2 cytokines exert reciprocal control over IFNproduction. IL-4 treatment during stimulation of human peripheral lymphocytes significantly elevated IFN1 transcription and secretion. However, pDCs were not directly responsive to IL-4. Using depletion and reconstitution experiments, we showed that IL-4– responsive monocytes are an intermediary cell, responding to IL-4 by elevating their secretion of IL-1 receptor antagonist (IL-Ra); this IL-1Ra acts on pDCs to elevate their IFN1 output. Thus, our experiments revealed a novel mechanism for regulation of both IFN1 production and pDC function, and suggests an expanded immunomodulatory role for Th2-associated cytokines. Phenotypic and Functional Comparison between Flask Adherent and Magnetic Activated Cell Sorted Monocytes Derived Dendritic Cells

Background: Generation of an effective dendritic cell (DC) based cancer vaccine depends on appropriate differentiation of monocytes in vitro. Objective: To compare the effects of monocyte separation methods, flask adherence (Flask-DC) and magnetic activated cell sorting (MACS-DC), on phenotypic and functional characteristics of resultant DCs. Methods: DCs from healthy volunteers were generated ...

Background & Aim: Tolerance to the semi-allogenic fetal graft by the maternal immune system is a medical enigma that has stimulated investigations for a half of century. Several hypotheses have been proposed to explain the tolerance of mother to the fetus. The successful pregnancy is proposed and proved by many scientists to be a Th2 dominant phenomenon. This hypothesis is proved in most as...

Maturation State and Function of Monocyte Derived Dendritic Cells in Liver Transplant Recipients

Background: Dendritic cells (DCs) are potent antigen presenting cells for triggering of the immune reaction post transplantation. These cells are centrally involved in the initiation of T cell-dependent immune responses. Objective: To compare the level of DC maturation and function in liver transplant recipients with healthy controls. Methods: In this study, twelve peripheral blood samples we...

Maturation of human iDCs by IL-18 plus PGE2, but not by each stimulus alone, induced migration toward CCL21 and the secretion of IL-12 and IFN-γ.

Dendritic cells (DCs) are potent antigen-presenting cells that initiate the primary immune response and whose functional properties in vivo depend on the maturation stimulus. We describe the functional properties of human monocyte-derived DCs after the maturation of immature DCs (iDCs) for 2 days with LPS (100 ng/ml), PGE2 (1 μg/ml), CD40L (1 μg/ml) or IL-18 (200 ng/ml) and with CD40L+PGE2 and ...

Interferon-gamma switches monocyte differentiation from dendritic cells to macrophages.

Human monocytes differentiate into dendritic cells (DCs) or macrophages according to the nature of environmental signals. Monocytes stimulated with granulocyte-macrophage colony-stimulating factor (GM-CSF) plus interleukin 4 (IL-4) yield DCs. We tested here whether interferon-gamma (IFN-gamma), a potent activator of macrophages, may modulate monocyte differentiation. Addition of IFN-gamma to IL...
