Cell-wall-bound lytic activity in CMoreT/a /wsca; function and characterization of an endo-mannanase

چکیده
A cell-wall-degrading activity was solubilized from young cells and from mother cell walls of CMo/W/a /w^ca by treatment with L i C l . The cytoplasmic enzyme hexokinase was not detectable in these extracts. The LiCl-solubilized activity increased in the cell cycle parallel to the release of autospores. The enzyme was purified on a chromatofocusing column followed by gel filtration. Sodium dodecyl sulfate/polyacryl amide gel electrophoresis of the purified enzyme revealed a molecular weight of 44 kDa, whereas gel filtration indicated a molecular weight of 25 kDa. Cell-wall-lytic activity and /M,4-mannanase activity coeluted in gel filtration and were separated from /?-D-fucosidase activity. The enzyme degraded isolated cell walls and ivory nut mannan primarily to oligosaccharides with an estimated degree of polymerisation ^ 6 . The soluble degradation products of the cell wall consisted of 92-96% mannose and 4-8% glucose. It is concluded that the cell-wall-lytic activity is caused by an endo-mannanase. In vivo, this enzyme probably degrades the mother cell wall and, after autospore release, remains bound to it as well as to the surface of the daughter cells by ionic forces. The identity of this bound enzyme with a soluble wall-degrading enzyme previously obtained from mother cells is discussed.

Cell wall-lytic activity in (j&Awn&Wa /wsca
The soluble fraction of homogenates of synchronous CMorg#a /w^ca was tested for carbohydrate-lyzing activities. With isolated cell walls and /?-l,4-mannan or carboxymethyl cellulose as substrates, a sharp increase in activity occurred shortly before release of the daughter cells followed by a decline during release. The lytic activities were partially purified by ammonium sulphate precipitation...
متن کاملMolecular cloning and biochemical characterization of an endo-β-mannanase gene from soybean for soybean meal improvement.
Soybean meal is the most commonly used protein source in animal feeds. Among the undesirable attributes of soybean meal is the high level of β-mannan, which was determined to be detrimental to the growth performance of animals. β-Mannan is a type of hemicellulose in the plant cell wall and can be hydrolyzed by endo-β-mannanase. The goal of this study is to isolate and characterize an endo-β-man...
متن کاملPRODUCTION, RELEASE AND THERMAL CHARACTERIZATION OF CELLULOLYTIC ENZYME FROM CELLULOMONAS sp. STRAIN "0"
Cellulase production by a Cellulomonas sp., isolated in 1985 from forest humus soil along the border of the Caspian Sea in Iran, was investigated. This strain secreted endo-and exo-cellulases in the culture medium, but some of the enzymes produced remained cell membrane bound. Cell bound enzymes were released by various treatments. The highest amount of endo-glucanase (up to 95%) and exo-gl...
متن کاملThe emergence of embryos from hard seeds is related to the structure of the cell walls of the micropylar endosperm, and not to endo-beta-mannanase activity.
BACKGROUND AND AIMS Seeds of carob, Chinese senna, date and fenugreek are hard due to thickened endosperm cell walls containing mannan polymers. How the radicle is able penetrate these thickened walls to complete seed germination is not clearly understood. The objective of this study was to determine if radicle emergence is related to the production of endo-beta-mannanase to weaken the mannan-r...
متن کاملA non-modular endo-beta-1,4-mannanase from Pseudomonas fluorescens subspecies cellulosa.
Pseudomonas fluorescens subsp. cellulosa when cultured in the presence of carob galactomannan degraded the polysaccharide. To isolate gene(s) from P. fluorescens subsp. cellulosa encoding endo-beta-1,4-mannanase (mannanase) activity, a genomic library of Pseudomonas DNA, constructed in lambda ZAPII, was screened for mannanase-expressing clones using the dye-labelled substrate, azo-carob galacto...
