Labelling of 6-iodocholesterol in melt.

It is well known that isotopic exchange of iodine in melt is a good approach to iodine labelling when the exchange in solution can not conveniently be achieved''. The advantage of this method is very efficient and rapid labelling technique, and the ease of preparation for labelling without addition or removal of solvent. However, it essentially required that the compound of interest is to be stable at the molten state. Another approach has been reported for iodine labelling of compounds melt with decomposition by using an unreactive solvent with a low melting point such as acetamide2'. Recently a melt method has been also adapted to nonisotopic exchange such as the I-for-Br and the F-for-Br exchange5',s'. The radioiodine analogue of 6-iodocholesterol (CL-6-I), synthesized by Liu Bo-Li, et al.,7' has been used clinically as a new diagnostic agent for adrenal gland in China. This labelled compound was prepared by exchange of unlabelled compound with radioactive iodine using nbutanol as a solvent. However, longer reaction time was necessary (90% exchange after 3 hours at refluxing temperature).
