چکیده

The effect of salts on the rate of penetration of dye into living cells has been studied by various investigators I by comparing the rate in dye solution containing salt with that in dye solution containing no salt, but it is not possible to determine from their results whether the salt acts on the protoplasm or on the dye or on both. The writer therefore has suggested a method ~,3 by which we are able to separate the effect of salt on the protoplasm from that on the dye. This is done as follows: (1) Cells are placed for a given time in a salt solution, after which they are placed in dye solution containing no salt. The rate of penetration of dye in the case of such cells is compared with that of the control (cells which have not been previously exposed to the salt solution). This gives the effect of salt on the protoplasm since it has previously been shown 2,3 that there is no error due to salt adhering to the surface of the cell wall (which cannot be removed by wiping and washing). I t might be thought that an error could arise from the fact that salt might diffuse out from the cell when removed from the salt solution and placed in the dye solution

Mechanism of the Accumulation of Dye in Nitella on the Basis of the Entrance of the Dye as Undissociated Molecules

The rate of penetration of brilliant cresyl blue into the living cells of Nitella indicates that the dye enters only in the form of the undissociated molecule. At equilibrium the total concentration of the dye in the sap is proportional to the concentration of the free base in the outside solution.

Counteraction of the Inhibiting Effects of Various Substances on Nitella

When living cells of Nitella are first exposed to (1) phosphate buffer mixture, or (2) phosphoric acid, or (3) hydrochloric acid, or (4) sodium chloride, or (5) sodium borate, and are then placed in a solution of brilliant cresyl blue made up with a borate buffer mixture at pH 7.85, the rate of penetration of the dye into the vacuole is decreased as compared with the rate in the case of cells t...

STUDIES ON PENETRATION OF DYES WITH GLASS ELECTRODE V. WHY DOES AZURE B PElq'ETRATE MORE READILY THAN METHYLENE BLUE OR CRYSTAL VIOLET? BY MARIAN IRWIN

The results 1 from measurements made with the glass electrode and spectrophotometer show that brilliant cresyl blue penetrates as free base into the vacuoles of living cells of Nitella. The present paper describes similar experiments with azure B, methylene blue, and crystal violet. Since the methods for the condition of cells, extraction of the sap, penetration experiments, extraction of free ...

Iv. Penetration of Brilliant Cresyl Blue into Nitella Flexilis by Marian Irwin

A basic dye is usually obtained in the form of the salt some of which may be transformed in aqueous solution to free base (the m o u n t depending on the "apparent" dissociation constant of the dye and the pH value), and in the range of pH values generally employed with living cells (approximately between pH 5 and 9.5) both forms of dye must be considered. In this range methylene blue, for exam...
