An introduction to particle separations in zonal centrifuges.

چکیده
Zonal centrifugation, based either on sediInentation rate or on isopycnic banding in liquid density gradients, is useful for high-resolution separation of subcellular particles. However, the amoun t of material which may be separated is extremely limited when conventional centrifuges are employed. To increase the capacity and, in certain instances, the resolution of the method, the possibility of employing largevolume hollow rotors has been explored. These are either loaded and unloaded while spinning or are designed to allow the density gl'adient in the rotor to reorient from a rest position to spin orientation and back with minimal shear and disturbance. The principles of operation and construction of low. speed A series and intermediate-speed B series zonal rotors are reviewed.Nat Cancer Inst Monogr 21: 9-39 1966.

The Development of Low - Speed " A " Series
The evolution of a series of low-speed zonal centrifuge rotors for the separation of cells, bacteria, and subcellular particles visible in the light microscope {Rotors A-V, VI, IX, and XII) is described. All are dynamically loaded and unloaded and may be used for either rate-zonal or isopycnic-zonal separations. Transparent plastic end caps allow direct observation of the separations. The last ...
متن کاملPreclinical Evaluation of 68Ga-MAA from Commercial Available 99mTc-MAA Kit
99mTc-Macroaggregated Albumin (99mTc-MAA) has been used as a perfusion agent. This study described development of the 68Ga-MAA via commercially available kits from Pars-Isotopes Company as a 99mTc-MAA kit. 68Ge/68Ga generator was eluted with suprapure HCl (0.6 M, 6 mL) in 0.5 mL fractions. The two fractions with the highest 68GaCl3 activity were generally used for labeling purposes. After label...
متن کاملIsolation of Relaxing Particles from Rat Skeletal Muscles in Zonal Centrifuges
Supernatants of rat skeletal muscle homogenates were fractionated by differential centrifugation and by zonal centrifugation in sucrose density gradients. Cytochrome oxidase was employed as an enzymatic marker for locating mitochondria. The subcellular fractions were also assayed for their ability to prevent the ATP-induced contraction of myofibrils. Both the mitochondrial and microsomal fracti...
متن کاملComparison of different types of rotor for density-gradient techniques.
Centrifugal separations can be classified into three categories, namely differential pelleting, rate-zonal centrifugation and isopycnic centrifugation. Although differential pelleting is the most widely used technique, it has only a limited potential for providing the investigator with purified material. Thus only the last two techniques, both of which involve separations on density gradients, ...
متن کاملA NEW CEMENT ADDITIVE TO IMPROVE THE PHYSICAL PROPERTIES OF OIL WELL CEMENT AND TO ENHANCE ZONAL ISOLATION
The main purpose of cementing of the well is to provide effective zonal isolation for the entire life of wells. The standard cements cannot prevent loss of zonal isolation when the stress level in the well cement is severely fluctuated. Since elastic cements improve elastic properties, recently, a number of studies have been done in this area. For this purpose, a nanoparticle, named EX-RIPI, wa...
