Studies of the Acetyl Coenzyme A Synthetase Reaction
نویسنده
چکیده
Acetyl coenzyme A synthetase, which catalyzes the biosynthesis of acetyl coenzyme A from acetat,e, adenosine triphosphate, and coenzyme A, has previously been obtained in highly purified form from bovine heart mitochondria (1). The enzyme appeared to be fairly homogeneous because only a single protein peak was detected in two chromatographic systems and the pattern observed in the analytical ultracentrifuge showed only minor contamination. However, the preparative procedure was lengthy and difficult to reproduce. Now, crystalline acetyl-CoA synthetase has been obtained which shows a specific activity over twice that found previously. This report describes the improved fractionation procedure and certain properties of the purified enzyme including its molecular weight.

متن کاملPurification and properties of acetyl coenzyme A synthetase from bovine heart mitochondria.
Acetyl coenzyme A synthetase has been partially purified from many sources including yeast, bacteria, molds, plants, mammalian organs, and pigeon liver (5-11). Substrate amounts of this enzyme, partially purified by the method of Hele from beef heart mitochondria (lo), have been used for the isolation of acetyl adenylate from a reaction mixture containing all reactants except coenzyme A (4). Al...

The acyl adenylates participate in a variety of synthetase reactions, and their recent isolation from reactions containing substrate quantities of enzyme suggests that these compounds act as enzyme-bound intermediates (2-7). However, such evidence does not esclude the possibility, emphasized by Boyer, Mills, and Fromm, that the adenylate occurs only as a free side product that is in equilibrium...

The enzymes catalysing the initial stage of the 8-oxidation of fatty acids, the acyl-CoA synthetases, have been classified into four groups based on specificity. These are: the short-chain (acetyl-CoA synthetase; EC 6.2.1 .l), medium-chain (butyryl-CoA synthetase; EC 6.2.1.2) and the long-chain fatty acyl-CoA synthetase (acyl-CoA synthetase; EC 6.2.1.3), which are ATP-dependent and follow the r...
متن کاملPurification and properties of aminoacetone synthetase from beef liver mitochondria.
Aminoacetone synthetase from beef liver mitochondria was purified to homogeneity and shown to be a member of the pyridoxal 5'-phosphate-dependent family of enzymes. This enzyme catalyzes the condensation of glycine and acetyl-CoA to produce CO2, CoA, and the stable product aminoacetone. Bovine aminoacetone synthetase is a dimer (Mr 56,000) of identical subunits and contains 2 mol of pyridoxal p...
