The Aberrant Long Non-Coding RNA NEAT1 Affected Nrf2/HO-1 in the Regulation of Hepatocellular Carcinoma

Aim: NEAT1 (nuclear-enriched abundant transcript 1) is the lncRNA that upregulated and functions as a promoter in hepatocellular carcinoma. This study aims to investigate role of interacted with nuclear factor erythroid-derived 2-related factor2 (Nrf2)/ heme oxygenase (HO-1) signaling pathway HCC. Methods: The expression profiling HCC tissues were identified by microarray analysis quantitative real-time PCR. effects either Nrf2 or knockdown on proliferation, invasion, apoptosis oxidative stress-related enzymes evaluated using Milliplex Human Oxidative Stress Magnetic Bead Panel. Xenograft tumor assay was carried out confirm cells vivo. Results: positive-correlated cell lines. Moreover, we found under hypoxia/reoxygenation conditions, activity (CAT, GSH-Px, SOD) significantly lower than before treatment(P< 0.01). factors involved Nrf2/HO-1 different after H/R treatment compared pre-treatment which indicating important relation stress. Knockdown led inhibition proliferation invasion promotion apoptosis, accompanying down-regulation Ki67, Keap-1 HO-1 up-regulation ROS levels. These observations confirmed xenograft mouse models wherein knockout proliferate at slower rate wild-type cells. Conclusions: promotes tumorigenesis affecting stress further regulating pathway.
