Production of Fructooligosaccharides Using a Commercial Heterologously Expressed Aspergillus sp. Fructosyltransferase

The catalytic properties of Seqenzym® FT, a fungal fructosyltransferase heterologously expressed in yeasts, were investigated at temperature 55 °C and pH 5.5. initial rate measurements showed that the transfructosylation was only slightly inhibited by sucrose above concentration 1.5 M. A rather low level hydrolytic side activity observed even concentrations as 0.25 In progress curve experiments, mass yield fructooligosaccharides (FOS) reached maximum value 57% this concentration, although it dropped to about 35% later on. At high up 2 M, FOS approximately 63% conversion 90%. Although neither nor changed much on, reaction manifested gradual depletion shorter chain FOS, 1-kestose nystose, accumulation 1-β-fructofuranosyl nystose. degree polymerization through number fructosyl units grew from 2.3 87% 3.1 94%. Compared other commercial preparations, FT can better produce with higher polymerization.
