Abstract Geminiviruses are causal agents of devastating diseases in crops. have circular single-stranded (ss) DNA genomes that replicated the nucleus infected plant cell through double-stranded (ds) intermediates by replication machinery. Which host polymerase mediates geminiviral multiplication, however, has so far remained elusive. Here, we show subunits nuclear replicative polymerases ? and ? physically interact with geminivirus-encoded enhancer protein, C3, these required for viral replication. Our results suggest that, while is essential to generate dsDNA intermediate, synthesis new copies ssDNA genome, virus-encoded C3 may act selectively, recruiting over ? favour productive
