Mechanisms of toxicity mediated by neutrophil and eosinophil granule proteins

چکیده

Neutrophils and eosinophils are granulocytes which characterized by the presence of granules in cytoplasm. Granules provide a safe storage site for granule proteins that play important roles immune function granulocytes. Upon activation, diverse released from into extracellular space contribute to fight against infections. In this article, we describe both neutrophils able kill pathogens review their anticipated mechanism antimicrobial toxicity. It should be noted an excess protein release can lead tissue damage host resulting chronic inflammation organ dysfunction.

Activation of platelets by eosinophil granule proteins

Two of the four principal cationic proteins of the eosinophil granule, major basic protein (MBP) and eosinophil peroxidase (EPO), were shown to be platelet agonists. Both MBP and EPO evoked a dose-dependent nonlytic secretion of platelet 5-hydroxytryptamine in unstirred platelet suspensions even in the presence of 10 microM indomethacin. MBP also evoked secretion of platelet alpha granule and l...

Terminally Differentiating Eosinophils Express Neutrophil Primary Granule Proteins as well as Eosinophil-specific Granule Proteins in a Temporal Manner

Neutrophils and eosinophils, 2 prominent granulocytes, are commonly derived from myelocytic progenitors through successive stages in the bone marrow. Our previous genome-wide transcriptomic data unexpectedly showed that genes encoding a multitude of neutrophil primary granule proteins (NPGPs) were markedly downregulated during the end period of eosinophilic terminal differentiation when cord bl...

Post-translational tyrosine nitration of eosinophil granule toxins mediated by eosinophil peroxidase.

Nitration of tyrosine residues has been observed during various acute and chronic inflammatory diseases. However, the mechanism of tyrosine nitration and the nature of the proteins that become tyrosine nitrated during inflammation remain unclear. Here we show that eosinophils but not other cell types including neutrophils contain nitrotyrosine-positive proteins in specific granules. Furthermore...

Mechanisms of granule-mediated cytotoxicity

The members of Judy Lieberman’s laboratory study cytotoxic T lymphocytes and their role in antiviral immunity. In addition to deciphering the cell death pathway induced by granzyme A, they are interested in how CD8þ T-cell function is regulated, especially in the setting of chronic infection and with a particular emphasis on HIV. They also try to apply their understanding to immunotherapy and v...

Eosinophil granule proteins expressed in ocular cicatricial pemphigoid.

BACKGROUND Blister formation and tissue damage in bullous pemphigoid have been attributed to the release of eosinophil granule proteins--namely, to eosinophil derived cationic protein (ECP) and major basic protein (MBP). In the present investigation these eosinophil granule proteins were studied in the conjunctiva of patients with ocular cicatricial pemphigoid (OCP). METHODS Conjunctival biop...
