Visualization of penicillin-binding proteins during sporulation of Streptomyces griseus.
We used fluorescein-tagged beta-lactam antibiotics to visualize penicillin-binding proteins (PBPs) in sporulating cultures of Streptomyces griseus. Six PBPs were identified in membranes prepared from growing and sporulating cultures. The binding activity of an 85-kDa PBP increased fourfold by 10 to 12 h of sporulation, at which time the sporulation septa were formed. Cefoxitin inhibited the int...
متن کاملInduction of beta-lactamase and penicillin-binding proteins in Escherichia coli by introduction of Streptomyces DNA.
Introduction of hybrid plasmids, which were constructed by ligation of pCR1 or pMN1 vector plasmid and SalI restriction endonuclease cleaved segments of Streptomyces cacaoi chromosome, resulted in the production of new beta-lactamase and penicillin-binding protein in Escherichia coli. The beta-lactamase and penicillin-binding protein were not from S. cacaoi but rather induced by the plasmids. C...
متن کاملDiscovering Lethal Proteins in Protein Interaction Networks
This chapter introduces state-of-the-art computational methods which discover lethal proteins from Protein Interaction Networks (PINs). Lethal proteins are an interesting subject in understanding the minimal condition for cellular development and survival. A dysfunctional research subject or absence of a lethal protein would result in fatality of the cell. Biological experiments have been condu...
متن کاملRecognition and characterization of Erythropoietin binding-proteins in the brain of mice
Objective(s): Erythropoietin (EPO), is a 34KDa glycoprotein hormone, which belongs to type 1 cytokine superfamily. EPO involves in erythrocyte maturation through inhibition of apoptosis in erythroid cells. Besides its main function, protective effects of EPO in heart and brain tissues have been reported. EPO has a critical role in development, growth, and homeostasis of brain. Furthermore EPO h...
متن کاملBINDING OF NAPROXEN TO SERUM PROTEINS IN PATIENTS WITH LIVER CIRRHOSIS AND HEALTHY INDIVIDUALS
A three-fold decrease of the in vitro ability of serum proteins to bind naproxen was shown in patients with cirrhosis of the liver compared to healthy individuals. This decrease was caused by quantitative changes in serum proteins in the sera of patients with liver damage. Lower doses of naproxen are therefore suggested in the treatment of patients with liver dysfunction.
