Developing T cells form an immunological synapse for passage through the ?-selection checkpoint

چکیده
The ?-selection checkpoint of T cell development tests whether the has recombined its genomic DNA to produce a functional receptor ? (TCR?). Passage through requires nascent TCR? protein mediate signaling pre-TCR complex. In this study, we show that developing cells at establish an immunological synapse in vitro and situ, resembling mature cell. is dependent on two key pathways known be critical for transition beyond checkpoint, Notch CXCR4 signaling. situ analyses indicate promotes passage checkpoint. Collectively, these data regulate formation synapse. This platform integrates cues from Notch, CXCR4, MHC thymic stromal allow

The Immunological Synapse T. Klemmensen: The Immunological Synapse
The induction of a proper adaptive immune response is dependent on the correct transfer of information between antigen-presenting cells (APCs) and antigen-specific T cells. Defects in information transfer may result in the development of diseases, e.g. immunodeficiencies and autoimmunity. A distinct 3-dimensional supramolecular structure at the T cell/APC interface has been suggested to be invo...
متن کاملImmunological synapse arrays: patterned protein surfaces that modulate immunological synapse structure formation in T cells.
T cells are activated by recognition of foreign peptides displayed on the surface of antigen presenting cells (APCs), an event that triggers assembly of a complex microscale structure at the T cell-APC interface known as the immunological synapse (IS). It remains unresolved whether the unique physical structure of the synapse itself impacts the functional response of T cells, independent of the...
