چکیده
Incorporation of membrane proteins into reconstituted lipid membranes is a common approach for studying their structure and function relationship in native-like environment. In this work, we investigated fluorescence properties liposome-reconstituted major light-harvesting complexes plants (LHCII). By utilizing liposome labelling with the fluorescent dye molecules single-molecule microscopy techniques, were able to study truly LHCII compare them bulk measurements liposome-free aggregates bound surface. Our results showed that lifetime obtained single correlated. The lifetimes shorter than LHCII. case LHCII, dependence on protein density reminiscent concentration quenching. size was not significant. demonstrated quenching can be induced by – interactions membranes, most likely occurring via same mechanism as photoprotective non-photochemical vivo.

Enhancement and quenching of single-molecule fluorescence.
We present an experimental and theoretical study of the fluorescence rate of a single molecule as a function of its distance to a laser-irradiated gold nanoparticle. The local field enhancement leads to an increased excitation rate whereas nonradiative energy transfer to the particle leads to a decrease of the quantum yield (quenching). Because of these competing effects, previous experiments s...
متن کاملSingle Molecule Spectroscopy of Monomeric LHCII: Experiment and Theory
We derive approximate equations of motion for excited state dynamics of a multilevel open quantum system weakly interacting with light to describe fluorescence-detected single molecule spectra. Based on the Frenkel exciton theory, we construct a model for the chlorophyll part of the LHCII complex of higher plants and its interaction with previously proposed excitation quencher in the form of th...
متن کاملNanomolar oligomerization and selective co-aggregation of α-synuclein pathogenic mutants revealed by single-molecule fluorescence
Protein aggregation is a hallmark of many neurodegenerative diseases, notably Alzheimer's and Parkinson's disease. Parkinson's disease is characterized by the presence of Lewy bodies, abnormal aggregates mainly composed of α-synuclein. Moreover, cases of familial Parkinson's disease have been linked to mutations in α-synuclein. In this study, we compared the behavior of wild-type (WT) α-synucle...

The ever-improving time and space resolution and molecular detection sensitivity of fluorescence microscopy offer unique opportunities to deepen our insights into the function of chemical and biological catalysts. Because single-molecule microscopy allows for counting the turnover events one by one, one can map the distribution of the catalytic activities of different sites in solid heterogeneo...
متن کاملSingle-molecule fluorescence spectroscopy of protein folding.
Single-molecule spectroscopy is an important new approach for studying the intrinsically heterogeneous process of protein folding. This Review illustrates how different single-molecule fluorescence techniques have improved our understanding of mechanistic aspects in protein folding, exemplified by a series of recent experiments on a small protein.
